Diagnosis, Clinical and Molecular Delineation of Human Plasmodium Species from Mangaluru, Southwest India

Malaria is a global threat and never-ending battle without appropriate identification differentiation of the parasite species. This work compared diagnostic methods including thick film microscopy technique, quantitative buffy coat, polymerase chain reaction. The inaccuracy species determination by consequent treatment regime underlines necessity to upgrade routine with molecular techniques. In study, 436 samples were collected; venous blood was processed for coat technique followed classical Giemsa staining thin smears nested Polymerase Chain Reaction (nPCR) genus-specific region Plasmodium targeting 18S rDNA species-specific identification. Of screened malaria, results in PCR showed 78.7% (100/127) be P. vivax, 4.8% (6/127) as falciparum 16.5% (21/127) mixed infection (P. vivax + falciparum). prevalence malaria 0.29, there good concordance between detecting (Kappa:0.77). our investigation, TFM exhibited sensitivity 97.7% specificity 100% detection QBC. Clinical parameters- thrombocytopenia anemia, this study. A positive association observed (p<0.05), but anemia remains unclear. Primer cross-reactions also primer sequence ovale knowlesi, sequencing confirmed it study phylogeny paved new way analyzing relatedness sequences.
